不 is a word that standfor negative, no, or not.
When we ask 在不在,direct english translation would be: there, not there.
And the answer will be either 在 (there), or 不在 (not there).
Same rules apply to other similar questions.
"Do you know how to ________?" is a question asking whether a person know how to or not know how to do something.
